Genome-wide identification of fungal GPI proteins.
-
A genome-wide search using a GPI-attachment consensus sequence identified 33 putative GPI-anchored proteins in S. pombe; SPBC1E8.05 (knh4) is among the predicted cell-surface GPI proteins.
"only 33 GPI candidates were identified"
Overexpression of cell-wall GPI-anchored proteins restores cell growth of N-glycosylation-defective och1 mutants in Schizosaccharomyces pombe.

SPBC1E8.05 (knh4) and pwp1 were identified as high-copy suppressors of the growth defect of N-glycosylation-defective och1-delta cells; both encode GPI-anchored proteins. Deletion of either gene is viable but confers sensitivity to beta-glucanase, indicating that Knh4 non-enzymatically supports beta-glucan on the S. pombe cell surface.

UniProt assigns O42970 to the Kre9/Knh1 family (Pfam PF10342 Kre9_KNH; InterPro IPR018466 Kre9/Knh1-like_N) with an N-terminal signal peptide, a serine-rich low-complexity/disordered region, predicted glycosylation, and a hydrophobic C-terminus; function is not experimentally determined (PE 2, evidence at transcript level).
