Gene Description

Lcp3 is a secreted structural protein of the Drosophila melanogaster larval chitin-based cuticle. The 112-residue precursor contains an N-terminal signal peptide and a Rebers–Riddiford cuticle-protein domain. Lcp3 contributes to the extracellular cuticular matrix produced by the larval epidermis; its expression and protein evidence are associated with the third-instar larval cuticle.

GO Term Evidence Action Reason
GO:0005214 structural constituent of chitin-based cuticle
IEA
GO_REF:0000002

Summary: Lcp3 has the more specific structural role in the larval chitin-based cuticle.
Reason: The broad structural annotation is biologically correct, but GO:0008010 captures the supported larval specificity. A0A0B4KEF3 and experimentally grounded reviewed Lcp3 P07188 encode the identical current 112-residue polypeptide and map to the same FlyBase gene. P07188 records direct protein sequencing from larval material and FlyBase IDA annotation to the larval-cuticle structural term; this is stronger evidence than the automated mapping alone.

The selected A0A0B4KEF3 sequence is identical to reviewed Lcp3 P07188: 112 of 112 residues match without gaps.

Summary: Lcp3 is a component of the extracellular larval cuticle.
Reason: The cuticle matrix location follows from the characterized Lcp3 protein, not just a secretion signal or an InterPro label. The selected sequence is identical to reviewed P07188, whose gene-level experimental annotation places it in the chitin-based extracellular matrix. The N-terminal signal peptide is consistent with this extracellular structural role.

External Prediction Reviews

These computational predictions are reviewed separately from the GOA annotation set used for this review. The assessments below are from this project and do not constitute official GO annotations or endorsement by GO/UniProt. They are not included in the existing annotation review above.

ProtNLM External predictions

ProtNLM2 emitted no GO or EC predictions for A0A0B4KEF3 in the frozen 2026-09-08 API snapshot. It emitted a protein name, a Secreted location, and the function text "Component of the rigid cuticle of the spider". The absence of GO output is an informative coverage omission: the current target is identical over all 112 residues to reviewed Lcp3 P07188 at the same FlyBase locus, which has experimental FlyBase annotations for GO:0008010 structural constituent of chitin-based larval cuticle and GO:0062129 chitin-based extracellular matrix. These are well-supported expected functions, not predictions emitted by ProtNLM. The separate function-description review supports the broad cuticle claim, leaves the rigid qualifier uncertain, and rejects the spider attribution. This record assesses missing GO output without assigning a VDCL score to an absent prediction or asserting a benchmark-wide recall value; the prediction-time input sequence and training-set membership remain unknown.
