Gene Description

GcvH2 is one of two KT2440 glycine cleavage H proteins. It is a small, non-catalytic carrier with a covalently attached lipoyl arm that accepts the glycine-derived aminomethyl group from GcvP, presents the intermediate to GcvT, and is subsequently reoxidized by a shared dihydrolipoyl dehydrogenase.

MARK AS OVER ANNOTATED
Summary: Cytoplasm is a generic parent of the more informative cytosol annotation already present.
Reason: Retain GO:0005829 as non-core location context instead of duplicating it with its parent.

KEEP AS NON CORE
Summary: A soluble cytosolic location is plausible for the bacterial carrier but is not a core function.
Reason: Retain the more specific of the two generic location annotations outside the core summary.

ACCEPT
Summary: GcvH2 is the lipoyl-bearing carrier component of the four-protein system.
Reason: The reviewed UniProt record explicitly identifies P, H, T, and L proteins as system components.

UNDECIDED
Summary: GcvH2 is a lipoylated carrier, but its precise role in the KT2440 lipoyl-transfer route has not been established.
Reason: Participation in a biological process does not require catalysis, and GcvH proteins can be obligatory intermediates in bacterial lipoyl relays. The covalent carrier state is certain, but available KT2440 evidence does not distinguish an active relay role from being only a LipA/LipB substrate.

ACCEPT
Summary: Intermediate shuttling by GcvH2 is required for this exact process.
Reason: UniProt describes transfer of the methylamine group from P protein to T protein on GcvH2.

NEW
Summary: This captures the covalently lipoylated carrier function omitted from current GOA.
Reason: The reviewed HAMAP-backed record states that GcvH2 binds one lipoyl cofactor covalently.
Supporting Evidence:

Binds 1 lipoyl cofactor covalently.

Core Functions

Covalently lipoylated H-protein carrier that shuttles the glycine-derived intermediate between the P, T, and L components.

Supporting Evidence:

Suggested Questions for Experts

Q: Does GcvH2 preferentially pair with its neighboring P2/T proteins or exchange freely with the first paralog set?

Suggested Experiments

Experiment: Reconstitute both P/H/T combinations and cross-combinations with each candidate E3 and compare turnover.

Type: in vitro pathway reconstitution
