Gene Description

UBE2F encodes a NEDD8-conjugating E2 that supports cullin-RING ubiquitin ligase activation. The short human product F8WDQ9 contains the N-terminal portion of UBE2F but lacks the catalytic cysteine and much of the UBC core present in the 185-residue reference product. Its expression, intracellular distribution and biochemical role are unresolved.

Q: Is the 101-residue F8WDQ9 product expressed endogenously, and does it bind E1, NEDD8 or cullin-ligase partners despite lacking the catalytic E2 cysteine?

The short UBE2F product lacks the catalytic cysteine and most of the E2 core. Intrinsic ubiquitin transfer is unsupported and contradicted by sequence and NEDD8 specificity; indirect ubiquitination involvement cannot be established for this product.

DOMAIN ARCHITECTURE MISMATCH
Review rationale: F8WDQ9 lacks the reference UBE2F catalytic Cys116 and a complete UBC core. Moreover, the characterized full-length enzyme conjugates NEDD8, not ubiquitin. A UBC-domain match does not establish ubiquitin-protein transferase activity.

UNC — Uncertain Review score: 1/2
Prediction method: ProtNLM2 · Version: UniProt API snapshot 2026-09-10 · file:human/UBE2F/UBE2F-protnlm-source.json
Review rationale: Full-length UBE2F supports CUL5-dependent ubiquitination through NEDD8 conjugation, so this biological process is not intrinsically incompatible with the gene. The 101-residue selected product lacks the catalytic E2 site and has no demonstrated assembly or regulatory function; its participation in protein ubiquitination is unresolved.
