Function
Receptor exemplar for agonist-driven Gs activation.
A compact Gs-coupled GPCR signaling module. An agonist-occupied GPCR activates heterotrimeric Gs, G alpha s stimulates adenylyl cyclase, cAMP accumulates and activates PKA, and PKA phosphorylates cytosolic and nuclear substrates including CREB. The module is grounded in GO:0007188. ADRB2 is anchored to PAINT GPCR nodes; downstream GNAS, ADCY5, PRKACA, and CREB1 are curated UniProt exemplars without PTN ancestry claims in this first pass.

Agonist-bound GPCR acts as a guanine nucleotide exchange factor for the heterotrimeric G protein, producing active G alpha s-GTP.
Receptor exemplar for agonist-driven Gs activation.
GTP-bound alpha subunit that stimulates adenylyl cyclase.
G alpha s activates membrane adenylyl cyclase to convert ATP to cAMP.
Enzyme tier that generates the cAMP second messenger.
cAMP binds PKA regulatory subunits, freeing catalytic subunits to phosphorylate substrates including CREB-family transcription factors.
cAMP-responsive kinase output of the pathway.
Nuclear transcriptional effector phosphorylated by PKA.
