Upstream membrane-cytoskeleton scaffold that promotes Hippo kinase activation in many contexts.
Hippo-YAP/TAZ signaling pathway module
A compact Hippo pathway module. Upstream polarity, junctional, mechanical, and stress inputs activate the MST1/2 kinase tier, which activates LATS1/2 kinases through scaffolded phosphorylation. Active LATS1/2 phosphorylate YAP/TAZ, causing cytoplasmic retention or degradation and suppressing TEAD-dependent growth programs. When the kinase cassette is inactive, YAP/TAZ enter the nucleus and coactivate TEAD transcription factors. The module is grounded in GO:0035329, with PAINT anchors for STK4/MST1 and LATS1 where local seed rows support the kinase-pathway roles.

Details
Connections
Cell-junction, polarity, cytoskeletal, and mechanical inputs converge on scaffolded activation of the MST1/2 kinase tier.
Annotons
Function
Proximal kinase tier that activates LATS1/2.
MST1/2 and scaffold proteins activate LATS1/2, the direct YAP/TAZ kinases.
Annotons
Function
Direct kinase tier that phosphorylates and inhibits YAP/TAZ.
LATS phosphorylation keeps YAP/TAZ out of the nucleus or promotes degradation. When Hippo kinase signaling is low, YAP/TAZ enter the nucleus and coactivate TEAD transcription factors.
Annotons
Locations
Regulated transcriptional coactivator controlled by LATS phosphorylation.
Locations
DNA-binding partner used by nuclear YAP/TAZ to drive growth-control genes.
